教育目標

This course will begin by introducing the fundamental biochemical concepts such as the stereochemical aspects of amino acids, peptides, proteins and nucleic acids, receptor-ligand interactions, enzyme-substrate interactions. More advanced topics such as complex enzyme and coenzyme mechanisms, specific molecular interactions of protein-ligand and DNA-small ligand will be taught. 2.0 credits are suggested for this course. 課程大綱 1. Structure and ionization properties of amino acids and polypeptides. Ionization equations and calculation of the net charge of polypeptides. 2. Secondary structures of proteins, the Ramachandran angles(dihedral angles) of peptides. Concepts of receptors, receptors ligand interactions, enzyme-substrate interactions. The Scatchard equation. 3. Importance of enzyme inhibitors and applications in modern drug developments. Derivation of the equations for competitive enzyme inhibitors 4. Enzyme catalytic principles and mechanisms: Aconitase, Carboxypeptidase, DNase I, Ribonuclease. Coenzyme and mechanisms: NADH (with X-ray structure of the enzyme-coenzyme complex), Biotin, Vitamin B12 5. DNA structure, major groove and minor groove figures. DNA-protein interactions, DNA-small ligand interactions. Restriction endonuclease mechanisms.
